用人淋巴细胞与~3H-地塞米松结合放射竞争抑制法测定糖皮质激素受体的数目,并观察口服地塞米松后受体数目的变化,以初步探讨激素和受体间作用的可能机理。 21名健康人各取静脉血30ml,EDTA抗凝,用Boyum不同比重的离心法去除血小板,分离淋巴细胞与多形核白细胞和红细胞。回收的淋巴细胞为62%~98%,混杂其中的多形核白细胞少于5%。取淋巴细胞混悬液(1.5~3.5)×10~6/ml与~3H-地塞米松混合,对照管中再加200倍地塞米松,两组浓度相等(1
The number of glucocorticoid receptor was determined by radioimmunoassay in combination with ~ 3H-dexamethasone and the number of receptors after oral administration of dexamethasone to investigate the possible mechanism of the interaction between hormone and receptor. Twenty-one healthy subjects received venous blood of 30 ml and EDTA anticoagulant. The platelets were removed by centrifugation with different specific gravity of Boyum, and lymphocytes, polymorphonuclear leukocytes and erythrocytes were isolated.
