目的:测定补阳还五汤全方的溶度峰及介电需量,为中药复方提取溶媒的选择提供理论与实验依据。方法:分别以溶度参数14.11~47.86,介电常数1~80的11种溶媒体系,在接近药材临界饱和溶媒体积V0,于25℃下浸提补阳还五汤粗粉,按《中国药典》2005年版附录浸膏得率测定项下测定表观溶解度。结果:各溶媒体系浸渍药材所需的临界饱和体积V0,分别为0.210,0.310,0.490,0.360,0.770,0.930,0.860,0.920,1.080,1.000,1.140 mL.g-1;各溶媒体系最大表观溶解度分别为114.0,101.3,73.4,109.4,210.7,295.0,501.4,437.0,355.6,423.1,210.6 g.L-1,从而得出补阳还五汤粗粉以甲醇-水(68∶32)浸渍时,其表观溶解度最大,出现溶度峰。对应的介电需量约为47.5。结论:中药及复方表观溶解度的大小决定于所提溶媒的介电常数;并有稳定的介电需量与之对应。
Objective: To determine the solubility peak and dielectric requirement of Buyang Huanwu Decoction in all aspects, and to provide theoretical and experimental evidence for the selection of Chinese herbal compound extraction solvent. METHODS: Eleven kinds of solvent media with solubility parameters of 14.11 to 47.86 and dielectric constants of 1 to 80 were used to extract Bu Yang Huan Wu Decoction coarse powder at a temperature of 25°C, close to the critical saturated media volume V0 of medicinal materials, according to the Chinese Pharmacopoeia. 》2005 Apparent Determination of Apparent Solubility in Appendix A Yield Determination. RESULTS: The critical saturated volume V0 required for impregnating each drug medium was 0.210, 0.310, 0.490, 0.360, 0.770, 0.930, 0.860, 0.920, 1.080, 1.000, 1.140 mL.g-1, respectively; The solubilities were 114.0, 101.3, 73.4, 109.4, 210.7, 295.0, 501.4, 437.0, 355.6, 423.1, 210.6 gL-1, respectively, and it was found that the Buyang Huanwu Tang coarse powder was impregnated with methanol-water (68:32). Its apparent solubility is the largest and there is a solubility peak. The corresponding dielectric requirement is about 47.5. Conclusion: The apparent solubility of traditional Chinese medicine and compound is determined by the dielectric constant of the proposed solvent; it also corresponds to a stable dielectric requirement.
