目的探讨半枝莲提取物(SBE)诱导人大肠癌细胞系(SW480、LoVo、HT-29)的凋亡作用及其可能机制。方法体外培养SW480、LoVo、HT-29细胞,以药物康复新、露蜂房作对照,应用MTT法和流式细胞术检测不同浓度SBE处理三种细胞48h后的抑制率、细胞凋亡率及周期变化;RT-PCR检测SBE处理后凋亡相关基因Bcl-2和Caspase-3mRNA表达。结果与康复新、露蜂房比较,SBE对三种细胞的抑制率和细胞凋亡率均呈剂量依赖性显著升高(P<0.01);S期细胞含量增多,出现S期阻滞;Caspase-3 mRNA表达亦显著上升,Bcl-2表达明显下降(P<0.01)。结论SBE具有诱导大肠癌细胞系凋亡和抑制增殖作用,作用强于康复新和露蜂房提取物。其机制可能与Caspase-3基因表达的上调及Bcl-2基因表达下降有关。
Objective To investigate the apoptosis of human colorectal cancer cell lines (SW480, LoVo, HT-29) induced by Scutellaria barbata extract (SBE) and its possible mechanism. Methods The SW480, LoVo and HT-29 cells were cultured in vitro. Drug rehabilitation new and honeycomb were used as control. MTT assay and flow cytometry were used to detect the inhibitory rate, apoptosis rate and cycle of the three kinds of cells treated with different concentrations of SBE for 48h. The expression of Bcl-2 and Caspase-3mRNA in the apoptosis-related genes after SBE treatment were detected by RT-PCR. Results Compared with the new and exposed hives, the inhibitory rate and apoptosis rate of SBE in three kinds of cells were increased in a dose-dependent manner (P <0.01). The content of S phase cells increased and S phase arrest. Caspase- 3 mRNA expression was also significantly increased, Bcl-2 expression was significantly decreased (P <0.01). Conclusion SBE can induce the apoptosis and inhibit the proliferation of colorectal cancer cell lines, and its effect is stronger than that of the recovered compound. The mechanism may be related to the up-regulation of Caspase-3 gene expression and the decrease of Bcl-2 gene expression.
