目的 研究caspase 3在粒细胞集落刺激因子 (G CSF)和粒 巨噬细胞集落刺激因子 (GM CSF)调节白血病细胞系NB4细胞凋亡过程中的变化及其意义。方法 利用细胞形态学、流式细胞仪分析及DNA片段化比率测定 ,检测G CSF及GM CSF对NB4细胞凋亡的影响 ,应用荧光分光光度法检测caspase3活性变化 ,并利用AC DEVD CHO进行caspase 3抑制试验。结果 G CSF能诱导NB4细胞凋亡 ,而GM CSF则无此作用 ;G CSF诱导的细胞凋亡过程中caspase 3活性明显升高 ,而AC DEVD CHO能有效地阻断G CSF诱导的细胞凋亡。结论 G CSF及GM CSF对NB4细胞的凋亡有着不同的调节作用 ,G CSF通过激活caspase 3而诱导白血病细胞的凋亡 ,caspase 3在细胞凋亡中起重要作用。
Objective To investigate the changes and significance of caspase 3 in regulating the apoptosis of leukemia cell line NB4 cells induced by granulocyte colony-stimulating factor (G CSF) and granulocyte macrophage colony-stimulating factor (GM CSF). Methods Cell morphology, flow cytometry analysis and DNA fragmentation ratio were used to detect the effect of G CSF and GM CSF on apoptosis of NB4 cells. The activity of caspase 3 was detected by fluorescence spectrophotometry, and AC DEVD CHO was used to perform caspase 3 assay. Inhibition test. Results G CSF could induce apoptosis of NB4 cells, while GM CSF did not. Gappa-CSF induced apoptotic increase in caspase 3 activity, whereas AC DEVD CHO effectively blocked G CSF-induced apoptosis. . Conclusion G CSF and GM CSF have different regulation effects on apoptosis of NB4 cells. G CSF induces apoptosis of leukemic cells through activation of caspase 3, and caspase 3 plays an important role in apoptosis.
