目的观测水通道蛋白在慢性应激肝郁气滞证模型大鼠组织中的表达,研究逍遥丸对其表达的影响。方法 30只大鼠随机分为正常组、模型组及中药组,采用慢性应激抑郁模型复制大鼠肝郁气滞证;中药组造模第7天始每天1次逍遥丸灌胃。以免疫组化研究AQP4及AQP3、AQP8在大鼠海马及胃肠中的表达。结果 AQP4在大鼠海马的表达无明显变化。三组中,中药组结肠黏膜中AQP3表达下调,三组平均光密度分别为(0.16±0.05),(0.21±0.07),(0.13±0.02)(F:1.039,P<0.05);模型组胃肠黏膜中AQP8表达上调,三组胃黏膜的平均光密度分别为(0.15±0.01),(0.19±0.04),(0.26±0.07)(F:8.591,P<0.05);三组小肠黏膜的平均光密度分别为(0.16±0.01),(0.21±0.04),(0.16±0.01),结肠黏膜的平均光密度分别为(0.16±0.01),(0.22±0.04),(0.15±0.03),与正常组、中药组经统计学处理有差异(F:3.219,P<0.05;F:8.821,P<0.01),中药组小肠与结肠黏膜中AQP8平均光密度与模型组比较,差异有统计学意义(P<0.01)。结论肝郁气滞证大鼠胃肠黏膜AQP8则表达上调,逍遥丸可以下调其在肠黏膜中的过度表达。
Objective To observe the expression of aquaporin (AQP) in chronic liver-Qi stagnation syndrome model rats, and to study the effect of Xiaoyao Pill on its expression. Methods Thirty rats were randomly divided into normal group, model group and traditional Chinese medicine group. Chronic stress depression model was used to replicate liver qi stagnation syndrome in rats. The Chinese herb group was administered with Xiaoyao Pill once daily on the seventh day. The expression of AQP4, AQP3 and AQP8 in the hippocampus and gastrointestinal tract of rats was detected by immunohistochemistry. Results The expression of AQP4 in rat hippocampus had no significant change. In the three groups, the expression of AQP3 in colonic mucosa of Chinese herbal medicine group was down-regulated. The average optical density of the three groups were (0.16 ± 0.05), (0.21 ± 0.07) and (0.13 ± 0.02) respectively (F: 1.039, The mean optical density of gastric mucosa in the three groups was (0.15 ± 0.01), (0.19 ± 0.04) and (0.26 ± 0.07), respectively (F: 8.591, P <0.05) The optical density of the colonic mucosa were (0.16 ± 0.01), (0.21 ± 0.04) and (0.16 ± 0.01), respectively. The average optical density of colonic mucosa were (0.16 ± 0.01), (0.22 ± 0.04) and (0.15 ± 0.03) (F: 3.219, P <0.05; F: 8.821, P <0.01). The mean optical density of AQP8 in small intestine and colonic mucosa in Chinese medicine group was significantly lower than that in model group (F: P <0.01). Conclusion Gastric mucosa AQP8 in rats with liver-qi stagnation syndrome is up-regulated, Xiaoyao Pill can down-regulate its overexpression in intestinal mucosa.
