目的解析CXCL12/CXCR4在人早孕期蜕膜腺上皮细胞存在共表达模式及其生物学作用。方法收集正常妊娠6~10周蜕膜组织由天佑医院妇产科门诊手术室提供,免疫细胞化学法检测蜕膜腺上皮细胞CXCL12/CXCR4的表达;ELISA法检测蜕膜腺上皮细胞培养上清CXCL12的分泌水平;MTT法分析CXCL12/CXCR4对蜕膜腺上皮细胞的生物学调节作用。结果免疫细胞化学发现人早孕期蜕膜腺上皮细胞同时表达CXCL12/CXCR4,且体外培养的蜕膜腺上皮细胞可自发分泌CXCL12;MTT法发现CXCL12处理后蜕膜腺上皮细胞的体外增殖活力无明显改变。结论 CXCL12/CXCR4在人早孕期蜕膜腺上皮细胞存在共表达模式。
OBJECTIVE: To analyze the coexpression pattern of CXCL12 / CXCR4 in human decidual glandular epithelial cells in early pregnancy and its biological role. Methods The deciduas from 6 to 10 weeks of normal pregnancy were collected from the outpatient department of gynecology and obstetrics in Tianyou Hospital. The expression of CXCL12 / CXCR4 was detected by immunocytochemistry. The expression of CXCL12 / CXCR4 in the decidual gland epithelial cells was detected by ELISA. The secretion of CXCL12 / CXCR4 was detected by MTT assay. The biological regulation of decidual glandular epithelial cells was analyzed by MTT assay. Results Immunocytochemistry showed that CXCL12 / CXCR4 was expressed in human decidual glandular epithelial cells in early pregnancy, and CXCL12 was secreted by human decidual glandular epithelial cells in vitro. MTT assay showed that the proliferation activity of decidual glandular epithelial cells after CXCL12 treatment was not significant change. Conclusion The expression of CXCL12 / CXCR4 in human early pregnancy decidual glandular epithelial cells coexist.
