目的观察华支睾吸虫分泌排泄抗原(ESA)刺激感染小鼠脾脏单个核细胞Th1/Th2细胞因子的动态变化。方法双抗体夹心ELISA法检测华支睾吸虫感染后不同时间小鼠脾细胞培养上清中IFN-γ、IL-4的动态变化。结果自感染后1w起IFN-γ的表达量开始上升,第4w达高峰(P<0.05),此后开始下降,至第12w水平接近正常。与IFN-γ不同,IL-4的表达自感染后第1w起持续升高,直至观察结束的12w(P<0.05)。结论初步结果提示华支睾吸虫感染后急性期转向慢性期的过程中Th1/Th2细胞因子的表达经历了由Th1优势应答向Th2优势应答的转变。
Objective To observe the dynamic changes of Th1 / Th2 cytokines in the splenic mononuclear cells of Clostridium testosteroni secreting excretion antigen (ESA). Methods Double antibody sandwich ELISA was used to detect the dynamic changes of IFN-γ and IL-4 in the culture supernatant of mouse spleen cells after infection with Clonorchis sinensis. Results The expression of IFN-γ began to increase from 1w after infection and peaked at 4w (P <0.05), and then began to decline. The level of IFN-γ began to decrease at the 12th week. In contrast to IFN-γ, IL-4 expression continued to increase from the 1st week after infection until 12 weeks after the end of observation (P <0.05). Conclusion The preliminary results suggest that Th1 / Th2 cytokines undergo a shift from Th1 predominant to Th2 predominant during the acute phase of C. sinensis infection.
