目的合成和分离一磺基酞菁锌(ZnPcS),并研究其对体外培养人急性白血病细胞(HL60细胞株)的光敏杀伤效应。方法采用高效液相色谱法分离纯化ZnPcS,以HL60细胞经药物作用48h红光照射5min后测定ZnPcS对HL60细胞的杀伤作用。结果ZnPcS浓度<20μg/ml对HL60细胞没有明显暗毒性,在红光照射下,10~20μg/mlZnPcS在体外能显著杀伤HL60细胞,并呈剂量的依赖性。结论ZnPcS对HL60细胞表现出很强的光敏杀伤作用,其作用机理可能是ZnPcS与细胞原生质膜相互作用产生高浓度单线态氧(1O2),引起细胞生物分子产生过氧化反应。
OBJECTIVE To synthesize and isolate ZnPcS, and study its phototoxicity on cultured human acute leukemia cells (HL60). Methods The ZnPcS was isolated and purified by high performance liquid chromatography (HPLC), and the killing effect of ZnPcS on HL60 cells was determined by irradiating HL60 cells for 48 h with red light for 5 min. Results ZnPcS concentration of <20μg / ml did not show any significant toxicity to HL60 cells. Under the irradiation of red light, 10 ~ 20μg / ml ZnPcS could significantly kill HL60 cells in vitro in a dose-dependent manner. Conclusions ZnPcS shows a strong phototoxicity on HL60 cells. The mechanism may be that ZnPcS interacts with the plasma membrane to generate high concentration of singlet oxygen (1O2), which causes the peroxidation of cell biomolecules.
