目的:提取并纯化放线共生放线杆菌表面相关物质。方法:厌氧环境下液体培养放线共生放线杆菌(Y4)72h,低温超速离心分离细菌,上清液经超滤浓缩后,以60%硫酸铵粗提,HPLC纯化,检测各峰细胞毒性以及蛋白质含量、总糖含量、内毒素含量,透射电镜观察。结果:透射电镜显示:液体培养72h多数细菌表面电子阻射区剥离;粗提物经HPLC显示一个蛋白主峰中含3个小蛋白峰,峰Ⅱ有细胞毒性,此峰为糖蛋白,内毒素含量极微。结论:提取、纯化毒素的主要毒力成份是细菌表面相关物质
Objective: To extract and purify the surface-related substances of Actinobacillus actinomycetemcomitans. Methods: Anaerobic conditions in liquid culture actinomycetemcomitans Actinobacillus actinomycetemcomitans (Y4) 72h, ultracentrifugation of bacteria at low temperature ultracentrifugation, the supernatant was concentrated by ultrafiltration, the crude extract was 60% ammonium sulfate, HPLC purified to detect peak cytotoxicity Protein content, total sugar content, endotoxin content and transmission electron microscopy were observed. Results: Transmission electron microscopy showed that most of the bacterial surface electron-exclusion zone was peeled off after 72 h of culture. The crude protein showed three small protein peaks and one peak Ⅱ cytotoxicity by HPLC. The peak was glycoprotein, endotoxin content Very slight. Conclusion: The main virulence component of toxins extracted and purified is bacterial surface-related substances
