目的探讨益母草注射液水溶性生物碱单体胆碱、葫芦巴碱对正常大鼠、豚鼠离体、正常豚鼠在体子宫活动的影响。方法从益母草注射液水溶性总生物碱部位中提取分离出单体胆碱、葫芦巴碱;采用正常(大鼠、豚鼠)离体子宫模型、未孕豚鼠豚鼠在体子宫模型观察其对子宫收缩的活动力、张力和频率的影响,用BL-420E生物机能系统记录子宫收缩。结果离体子宫试验表明,胆碱、葫芦巴碱3×10-5可显著降低大鼠正常离体子宫收缩最大值(P<0.05),显著增加豚鼠正常离体子宫活动力(P<0.05),显著降低子宫收缩频率(P<0.05);在体子宫试验表明,肌肉注射1mg/kg胆碱,能够显著降低给药后60~80min正常豚鼠子宫的收缩频率(P<0.05),显著增加给药后0~10min子宫收缩张力最大值(P<0.05或P<0.01),对子宫收缩活力,收缩张力均值及最小值无明显影响;肌肉注射1mg/kg葫芦巴碱,能够显著增加给药后20~40min正常豚鼠子宫的收缩活力(P<0.05),显著增加给药后20~90min子宫收缩张力最大值(P<0.05或P<0.01),对子宫收缩频率、收缩张力均值及最小值无明显影响。结论胆碱、葫芦巴碱具有兴奋正常豚鼠离体子宫和在体子宫平滑肌活动的作用。
Objective To investigate the effects of monosodium glutamate and triglyceride, a water-soluble alkaloid, on the activities of uterus in normal and guinea pigs in vitro and in vivo. Methods Monomeric choline and trigonelline were isolated and purified from the fraction of water-soluble alkaloids extracted from Leonurus polygalae injection. Uterine contractions were observed in uterus of non-pregnant guinea-pig guinea-pigs using normal (rat, guinea pig) The activity, tension and frequency of the impact, using the BL-420E biofeedback system to record uterine contractions. Results In vitro uterus test showed that choline and trigonelline 3 × 10-5 could significantly reduce the normal uterine contractions in rats (P <0.05), significantly increase the normal uterine activity in guinea pigs (P <0.05) , Significantly reduced the frequency of uterine contractions (P <0.05). In vivo uterine test showed that intramuscular injection of 1 mg / kg choline significantly reduced the contraction frequency of guinea pig uterus (P <0.05) The maximum value of uterine contractility tension (P <0.05 or P <0.01) at 0-10 min after drug administration had no significant effect on uterine contractile activity and mean and minimum value of contractive tension. Intramuscular injection of 1 mg / kg of trigonelline could significantly increase the contractile tension The contractile activity of uterus of normal guinea pigs at 20 ~ 40min (P <0.05) increased significantly (P <0.05 or P <0.01) between 20 ~ 90min after administration, Clearly affected. Conclusion Choline and trigonelline have the activity of excitating normal uterine guinea pig uterus and normal uterine smooth muscle.
