目的:探讨EB病毒编码的LMP1在体内鼻咽癌细胞中,是否可能通过影响鼻咽癌细胞增殖和凋亡的生物学过程,而在鼻咽癌的发展演进过程中起作用。方法:采用TUNEL法原位检测癌细胞的调亡程度,用免疫组化LSAB法检测LMP1、PCNA、p53和bcl-2蛋白表达Z分析LMP1的表达与癌细胞增殖、凋亡以及与p53、bcl-2表达的相关性。结果:LMP1的表达与否,与鼻咽癌细胞增殖活性的差异无显著意义,而与癌细胞凋亡程度之间的差异有显著意义,其表达与癌细胞凋亡程度呈正相关;LPM1的表达与p53和bcl-2的表达无相关性。结论:提示LMP1在体内鼻咽癌组织中可能有促使癌细胞凋亡的生物学作用,其机制可能是通过激活细胞毒性T淋巴细胞,增强后者对癌细胞的杀伤作用。LMP1对癌细胞增殖和凋亡的作用可能并不受p53及bcl-2表达的影响。
AIM: To investigate whether LMP1 encoded by Epstein-Barr virus (EB) can play a role in the development and progression of nasopharyngeal carcinoma in nasopharyngeal carcinoma cells through the biological processes that affect the proliferation and apoptosis of nasopharyngeal carcinoma cells. Methods: TUNEL method was used to detect the apoptosis of cancer cells in situ. The expression of LMP1, PCNA, p53 and bcl-2 was detected by immunohistochemical method. The expressions of LMP1, -2 expression of the correlation. Results: The expression of LMP1 was not significantly different from that of nasopharyngeal carcinoma cells, but significant difference was observed between the expression of LMP1 and the degree of apoptosis. The expression of LMP1 was positively correlated with the degree of apoptosis of the cancer cells. The expression of LPM1 No correlation with p53 and bcl-2 expression. CONCLUSION: It is suggested that LMP1 may have the biological role in promoting the apoptosis of human nasopharyngeal carcinoma in vivo. The mechanism may be through activating cytotoxic T lymphocytes and enhancing the killing effect of LMP1 on cancer cells. The effect of LMP1 on the proliferation and apoptosis of cancer cells may not be affected by the expression of p53 and bcl-2.
