目的:探讨八聚体结合蛋白4(Oct4)、细胞周期蛋白D1(Cyclin D1)在胃腺癌中的表达及临床意义,并研究两因子之间的关系。方法:应用免疫组织化学(En Vision TM)法分别检测Oct4、Cyclin D1在70例胃腺癌、50例癌前病变及40例黏膜慢性炎组织中的表达情况。结果:Oct4在胃腺癌、癌前病变及黏膜慢性炎组织中的阳性表达率分别为55.71%、22.00%、7.50%,3组之间表达差异有显著统计学意义(χ2=30.78,P<0.01);Cyclin D1在胃腺癌、癌前病变及黏膜慢性炎组织中的阳性表达率分别为57.14%、14.00%、0,3组之间表达差异有显著统计学意义(χ2=48.35,P<0.01)。胃腺癌组织中Oct4的表达与肿瘤直径、TNM分期、淋巴结转移有关(P<0.05),但与性别、年龄、肿瘤位置、组织学分化、浸润深度无关(P>0.05);胃腺癌组织中Cyclin D1的表达与TNM分期、淋巴结转移有关(P<0.05),但与性别、年龄、肿瘤直径、肿瘤位置、组织学分化、浸润深度无关(P>0.05)。癌组织Oct4表达与Cyclin D1表达呈正相关(r=0.57,P<0.01)。结论:胃腺癌Oct4、Cyclin D1的过表达促进了肿瘤细胞的无限制增殖,为肿瘤细胞转移提供了必要条件。
Objective: To investigate the expression and clinical significance of Oct4 and Cyclin D1 in gastric adenocarcinoma and to study the relationship between the two factors. Methods: The expressions of Oct4 and Cyclin D1 in 70 cases of gastric adenocarcinoma, 50 cases of precancerous lesions and 40 cases of chronic mucosal inflammation were detected by immunohistochemistry (EnVision). Results: The positive expression rates of Oct4 in gastric adenocarcinoma, precancerous lesions and chronic mucositis tissues were 55.71%, 22.00%, 7.50%, respectively. There was significant difference in the expression of Oct4 between the three groups (χ2 = 30.78, P <0.01) ). The positive rates of Cyclin D1 expression in gastric adenocarcinoma, precancerous lesions and chronic mucositis tissues were 57.14% and 14.00%, respectively. There was significant difference between the three groups (χ2 = 48.35, P <0.01) ). The expression of Oct4 in gastric adenocarcinoma was correlated with tumor diameter, TNM stage and lymph node metastasis (P <0.05), but had no correlation with gender, age, tumor location, histological differentiation and depth of invasion (P> 0.05) The expression of D1 was correlated with TNM stage and lymph node metastasis (P <0.05), but not with gender, age, tumor diameter, tumor location, histological differentiation, depth of invasion (P> 0.05). The expression of Oct4 in cancer tissues was positively correlated with the expression of Cyclin D1 (r = 0.57, P <0.01). Conclusion: Overexpression of Oct4 and Cyclin D1 in gastric adenocarcinoma promotes the unlimited proliferation of tumor cells and provides the necessary conditions for tumor cell metastasis.
