采用大鼠和犬,经连续3mon口服给药,对雷公藤微囊(TWH)的毒性进行了研究。5mg·kg-1TWH对大鼠和犬均不表现任何毒性作用,TWH15mg·kg-1对大鼠,10mg·kg-1对犬有一定的毒性作用,但停药后,可以恢复;高剂量下(大鼠35mg·kg-1,犬15mg·kg-1),大鼠血糖、血尿素氮及犬的碱性磷酸酶活性显著增高,犬血总蛋白及血白蛋白含量显著下降,病理组织学检查也表明大鼠及犬的心、肝、肾等多种脏器有明显损害,但上述毒性停药后基本可以恢复,也未发现有明显的延迟毒性,表明高剂量TWH所产生的毒性是可逆的。
Rats and dogs were used to study the toxicity of Tripterygium wilfordii microencapsulated (TWH) by continuous oral administration of 3mon. 5mg·kg-1TWH did not show any toxic effects on rats and dogs. TWH15mg·kg-1 had certain toxic effects on rats in 10mg·kg-1 but it could be restored after discontinuation; high dose (35 mg·kg-1 in rats, 15 mg·kg-1 in dogs), blood glucose, blood urea nitrogen, and alkaline phosphatase activity in dogs were significantly higher, and total protein and serum albumin levels in dogs decreased significantly. Histopathology Examination also showed that rats, dogs, and other organs, liver, kidney and other organs have obvious damage, but the above-mentioned toxicity can be basically restored after drug withdrawal, and no significant delayed toxicity was found, indicating that the toxicity of high-dose TWH is reversible.
