目的 探讨哮喘大鼠肺组织中白血病抑制因子 (leukemiainhibitoryfactor ,LIF)的表达及其意义。方法 制作卵蛋白致敏大鼠哮喘模型 ,采用RT -PCR和Western -blot方法观察LIF在致敏大鼠肺组织中的表达。结果 LIF蛋白表达量在哮喘组为 (40 832± 12 96 4 ) ,均较地塞米松组 (16 16 0± 2 10 8)和对照组 (2 3110± 80 18)升高 ,差异有显著性 (P <0 .0 1) ,地塞米松组与对照组之间无显著性差异 (P >0 .0 5 )。LIFmRNA的相对表达量哮喘组为 (0 .5 1± 0 .13) ,均较地塞米松组 (0 .18± 0 .0 5 )和对照组 (0 .2 4± 0 .0 2 )明显增高 ,差异有显著性 (P <0 .0 1) ,地塞米松组与对照组之间无显著性差异 (P >0 .0 5 )。结论 LIF在致敏哮喘大鼠模型肺组织中表达显著增高 ,LIF作为神经免疫调节因子 ,可能参与了哮喘的发病机制。
Objective To investigate the expression of leukemia inhibitory factor (LIF) in the lung tissue of asthmatic rats and its significance. Methods Ovalbumin-sensitized rat asthma model was made. The expression of LIF in lung tissue of sensitized rats was observed by RT-PCR and Western-blot. Results Compared with dexamethasone group (16 16 ± 2 10 8) and control group (31 31 ± 80 18), the expression of LIF protein in asthma group was (40 832 ± 12 96 4), the difference was significant (P <0.01). There was no significant difference between dexamethasone group and control group (P> 0.05). The relative expression of LIF mRNA in asthma group was (0.51 ± 0.13), which was significantly lower than that in dexamethasone group (0.18 ± 0.05) and control group (0.24 ± 0.0.2) (P <0.01). There was no significant difference between dexamethasone group and control group (P> 0.05). Conclusion The expression of LIF in the lung tissue of sensitized asthmatic rat model was significantly increased. LIF, as a neuroimmunoregulatory factor, may be involved in the pathogenesis of asthma.
