In DNA replication, the single-stranded binding protein SSB, together with the SV40 T antigen and topoisomerase, unwinds the DNA so that other replication proteins approach the replication site. It also has the effect of activating the polymerase alpha and delta subunits. The authors report that SSB is also involved in the excision and repair of nucleotides in an in vitro system. The authors tested the ability of the system to repair DNA by assaying the radioactivity of bound or uninjured circular plasmid DNA together with four deoxynucleotides and [alpha-32P] dATP in hela cell extracts.
