对南瓜多糖进行提取,分离纯化。通过离子交换层析和凝胶过滤层析得到P11、P12、P213个组分,分别以清除DPPH自由基、羟基自由基和还原能力为指标,研究南瓜粗多糖及其3个组分的抗氧化活性。试验结果表明:南瓜粗多糖、P12和P21清除DPPH自由基的IC50分别为5.496、8.908、3.153mg/mL,南瓜粗多糖、P11、P12和P21清除羟基自由基的IC50分别为4.251、1.191、7.655、5.221mg/mL,南瓜粗多糖、P11、P12和P21的FRAP值依次为0.182、0.062、0.082、0.400mmol/g。
Pumpkin polysaccharide extraction, separation and purification. P11, P12 and P213 fractions were obtained by ion-exchange chromatography and gel filtration chromatography. The DPPH radical scavenging activity, hydroxyl radical scavenging activity and reducing ability were used as indexes to study the antioxidant activity of pumpkin crude polysaccharide and its three components active. The results showed that IC50 of pumpkin crude polysaccharide, DP12 and P21 scavenging DPPH free radicals were 5.496, 8.908 and 3.153 mg / mL, IC50 of scavenging pumpkin polysaccharide, P11, P12 and P21 were 4.251,1.191 and 7.655 , 5.221mg / mL, squash polysaccharide, P11, P12 and P21 FRAP values were 0.182,0.062,0.082,0.400mmol / g.
