目的 研究左旋多巴 (L- dopa)的神经毒性及其神经保护方法。方法 用胎盼兰染色法检测 L- dopa和抗氧化剂还原型谷胱甘肽 (GSH)、NMDA受体拮抗剂 MK80 1对体外培养大鼠纹状体的神经元活力的影响。结果 L- dopa使大鼠纹状体神经元活力明显下降 (P<0 .0 1) ,GSH和 MK80 1可明显减轻 L- dopa引起的纹状体神经元活力下降 (P<0 .0 1)。结论 L- dopa可能通过氧化应激和兴奋 NMDA受体两个途径发挥其神经毒性。抗氧化剂或 NMDA受体拮抗剂与 L- dopa合用可能减轻 L- dopa的神经毒性。
Objective To study the neurotoxicity and neuroprotection of L-dopa. Methods The effects of L-dopa, GSH and NMDA receptor antagonist MK80 1 on the neuronal viability in rat striatum were detected by using fetal blue staining. Results L-dopa significantly decreased the viability of striatum neurons (P <0.01). GSH and MK80 1 significantly attenuated the decrease of striatal neuronal activity induced by L-dopa (P <0.01) ). Conclusion L-dopa may exert its neurotoxicity through both oxidative stress and excitatory NMDA receptors. Antioxidants or NMDA receptor antagonists in combination with L-dopa may reduce the neurotoxicity of L-dopa.
