目的:研究中药温化蠲痹方对胶原诱导性关节炎(CIA)大鼠外周血TNF-α、IL-1β的影响,探讨温化蠲痹方治疗CIA的作用机制。方法:选用健康雄性Wistar大鼠,随机分为造模组、正常对照组(10只,简称正常组),造模组采用牛Ⅱ型胶原乳剂于大鼠尾根部注射,建立CIA模型。再把造模组随机分为模型组(10只)、MTX组(10只)、温化蠲痹方组(10只,简称中药组)。正常组、模型组、中药组按22.9g/kg剂量分别灌胃生理盐水、温化蠲痹方,均每日1次,MTX组按0.78mg/kg剂量灌胃MTX混悬液,每周1次。给药30天后,运用ELISA法检测大鼠外周血TNF-α、IL-1β水平。结果:与模型组比较,治疗后中药组和MTX组大鼠关节炎指数(AI)评分明显降低(P<0.01);与治疗前比较,中药组和MTX组大鼠AI评分均明显降低(P<0.01)。与正常组比较,模型组大鼠外周血TNF-α、IL-1水平明显升高(P<0.01);与模型组比较,中药组和MTX组大鼠外周血TNF-α、IL-1水平明显降低(P<0.01),两组之间比较差异无显著性(P>0.05)。结论:CIA大鼠外周血TNF-α、IL-1水平升高,降低CIA大鼠外周血TNF-α、IL-1水平可能是中药温化蠲痹方治疗CIA的作用机制之一。
Objective: To study the effect of Wenhua Huibi Recipe on TNF-α and IL-1β in peripheral blood of rats with collagen-induced arthritis (CIA) and to explore the mechanism of action of Wenhua Wanbi recipe in treating CIA. Methods: Healthy male Wistar rats were randomly divided into model group and normal control group (n = 10, referred to as normal group). The model rats were injected with the tail of the rats with type Ⅱ collagen emulsion to establish CIA model. Then the model group was randomly divided into model group (n = 10), MTX group (n = 10), and Wenhua Fangbi group (n = 10). The normal group, the model group and the traditional Chinese medicine group were given physiological saline at a dose of 22.9g / kg and warmed Bibi prescription once daily. The MTX group was orally administered MTX suspension at a dose of 0.78mg / kg every week Times. Thirty days after administration, the levels of TNF-α and IL-1β in peripheral blood of rats were detected by ELISA. Results: Compared with the model group, the arthritis index (AI) scores of the Chinese medicine group and the MTX group were significantly decreased after treatment (P <0.01). Compared with the pretreatment group, the AI scores of the Chinese medicine group and the MTX group were significantly decreased <0.01). Compared with the normal group, the levels of TNF-α and IL-1 in the peripheral blood of the model group were significantly increased (P <0.01). Compared with the model group, the levels of TNF-α and IL-1 in the Chinese herbal medicine group and the MTX group (P <0.01). There was no significant difference between the two groups (P> 0.05). CONCLUSION: The levels of TNF-α and IL-1 in peripheral blood of CIA rats are increased, and the levels of TNF-α and IL-1 in peripheral blood of CIA rats may be one of the mechanisms of CClA treatment.
