目的半定量研究产前缺氧对24周龄新西兰兔颈动脉转化生长因子β/Smad3表达及血管形态学的影响。方法同期怀孕的新西兰母兔[足月,(30±1)d]随机分为常氧对照组21%O2(n=5)、缺氧组(12%O2,n=5),于妊娠第10天开始相应的干预、记录,至妊娠第29天结束,自然分娩。每窝随机留1只雄性新生兔饲养,共得10只雄性新生兔。母乳饲养6周后,再给予普通饲料喂养至24周。利用Westernblot、免疫组织化学技术检测颈动脉转化生长因子β/Smad3表达;利用HE染色光镜及透射电镜观察24周龄兔颈动脉形态学表现。结果产前缺氧下调24周龄兔颈动脉转化生长因子β1/Smad3表达(均P<0.05);HE染色光镜及透射电镜结果表明,产前缺氧诱发24周龄兔颈动脉早期动脉粥样硬化的特征表现。结论产前缺氧在转录后水平下调24周龄兔颈动脉转化生长因子β1/Smad3信号通路,诱发早期动脉粥样硬化表现。
Objective To semi-quantitatively study the effect of prenatal hypoxia on the expression of TGF-β / Smad3 and vascular morphology in the carotid artery of New Zealand rabbits at 24 weeks of age. Methods Pregnant New Zealand females at the same period (full term, (30 ± 1) d] were randomly divided into normoxic control group (21% O2) and hypoxia group (12% O2 n = 5) 10 days to start the appropriate intervention, record, to the end of the 29th day of pregnancy, natural childbirth. One male neonatal rabbit was randomly housed in each litter, and a total of 10 male newborn rabbits were obtained. After six weeks of breastfeeding, normal diet was fed for another 24 weeks. The expression of TGF-β / Smad3 in carotid arteries was detected by Western blot and immunohistochemistry. Morphology of carotid arteries in 24-week-old rabbits was observed by HE staining and transmission electron microscopy. Results Prenatal hypoxia down-regulated the expression of TGF-β1 / Smad3 in the carotid arteries of 24-week-old rabbits (all P <0.05). The results of light microscopy and transmission electron microscopy showed that prenatal hypoxia induced carotid artery atherosclerosis The characteristic features of sclerosis. Conclusion Prenatal hypoxia down-regulates the carotid artery TGF-β1 / Smad3 signaling pathway in 24-week-old rabbits at post-transcriptional level to induce early atherosclerosis.
