目的观察IgA肾病(IgA nephropathy,IgAN)患者与正常人的血清热聚合IgA1(aggregated IgA1,aIgA1)对足细胞增殖的影响。方法收集原发性IgAN患者及健康人的血清,利用层析法分离获得血清单体IgA1(monomeric IgA1,mIgA1),将mIgA1热聚合为aIgA1。利用浓度为0.25、0.5、1.2 mg/ml的患者与正常人的aIgA1分别刺激小鼠MPC5足细胞株,利用MTT法检测aIgA1对足细胞增殖的影响。结果正常组和患者组aIgA1刺激足细胞后细胞MTT吸光度的差异无统计学意义。刺激24 h组与刺激48 h组MTT吸光度的差异也无统计学意义。不同浓度aIgA1刺激组间MTT吸光度的差异有统计学意义P<0.05),相比于不加刺激的阴性对照组(SFM),1、2 mg/ml组的吸光度分别为其0.46和1.66倍。结论足细胞可能会直接对IgA1产生反应,aIgA1可以影响足细胞增殖;低浓度aIgA1抑制足细胞增殖,高浓度aIgA1促进足细胞增殖;患者与正常人的aIgA1对足细胞增殖的影响没有不同。
Objective To investigate the effect of aggregated IgA1 (aIgA1) on the proliferation of podocytes in patients with IgA nephropathy (IgAN) and normal controls. Methods Serum from patients with primary IgAN and healthy individuals were collected. Serum monomeric IgA1 (mIgA1) was isolated by chromatography, and mIgA1 was thermally polymerized into aIgA1. The effects of aIgA1 on podocyte proliferation were detected by MTT assay using aIgA1 of 0.25, 0.5 and 1.2 mg / ml respectively. Results There was no significant difference in MTT absorbance between aIgA1-stimulated podocyte and normal group. There was no significant difference in the MTT absorbance between stimulated 24 h group and stimulated 48 h group. The difference of MTT absorbance between different concentrations of aIgA1 stimulation group was statistically significant (P <0.05). Compared with unstimulated negative control group (SFM), absorbance of 1,2 mg / ml group was 0.46 and 1.66 times respectively. Conclusion Podocytes may directly respond to IgA1, aIgA1 may affect podocyte proliferation; aIgA1 at low concentrations inhibits podocyte proliferation, and aIgA1 at high concentrations promotes podocyte proliferation; there is no difference in the effect of aIgA1 on podocyte proliferation between normal and human subjects.
