目的采用生物信息和网络信息技术建立一个带有临床资料和代表性克隆基因的多功能乙型肝炎病毒(HBV)序列数据库及HBV耐药分析平台。方法平台的构建基于J2EE框架,支持Windows、Linux等操作系统,支持Oracle、Sqlserver、Mysql等数据库。整合了序列比对、进化分析、抗原表位分析、耐药变异分析和耐药分析等生物信息学功能。收录的HBV基因序列、克隆和相关临床信息来源于2007年7月-2009年6月解放军302医院诊治的6880例HBV感染患者。结果初步建立了该信息平台并进行了应用。平台收录了本课题组检测分析的HBV全基因组序列516条,HBV反转录酶(RT)/S或前核心启动子/前C功能基因或调控序列11 456条,代表性HBV基因组或基因克隆613条以及患者的临床资料,实现了HBV序列比对、基因型/亚型和血清型分型、S/C/P/X蛋白氨基酸序列预测、T细胞抗原表位分析、耐药分析、患者临床信息检索等功能。应用该数据平台分析我国大样本慢性HBV感染患者HBV耐药变异和基因型流行特点,表明功能实用可靠。结论 HBV序列和耐药分析信息平台的建立有助于充分利用我国患者的HBV序列和相关信息资源,服务于乙肝患者的临床耐药检测和HBV病毒学特性研究。
Objective To establish a multi-functional Hepatitis B virus (HBV) database with clinical data and representative cloning genes and bioinformatics analysis platform based on bioinformatics and network information technology. The method platform is based on J2EE framework, supports Windows, Linux and other operating systems, supports Oracle, Sqlserver, Mysql and other databases. Integration of bioinformatics functions such as sequence alignment, evolutionary analysis, epitope analysis, resistance mutation analysis and drug resistance analysis. The included HBV gene sequences, cloning and related clinical information are from 6880 cases of HBV infection diagnosed and treated by People’s Liberation Army 302 Hospital from July 2007 to June 2009. Results The information platform was initially established and applied. The platform included 516 HBV genome sequences, HBV reverse transcriptase (RT) / S or pre-core promoter / pre-C functional gene or regulatory sequence 11 456, representative HBV genomes or gene clones detected and analyzed by our group 613 and the clinical data of the patients to achieve HBV sequence alignment, genotyping / subtype and serotype typing, amino acid sequence prediction of S / C / P / X protein, T cell epitope analysis, drug resistance analysis, patients Clinical information retrieval and other functions. The application of the data platform to analyze the characteristics of HBV resistance and genotypic prevalence in patients with chronic HBV infection in large sample in China indicates that the function is practical and reliable. Conclusion The establishment of HBV sequence and drug resistance analysis information platform will help make full use of HBV sequences and related information resources of patients in our country and serve as a reference for clinical drug resistance testing and HBV virological research in hepatitis B patients.
