目的探讨大鼠阿维菌素经口给药对脑细胞的细胞毒性。方法大鼠经口给药,观察急性中毒发生过程,取大鼠脑组织做组织病理学检查,并用MTT法测定细胞毒性。结果急性中毒发生后可见神经毒性症状,组织病理学检查可见脑细胞变性或坏死的改变;MTT结果 100mg/kg组与对照组比较,差异有统计学意义(P<0.05)。结论在本试验条件下,虽有血脑屏障的作用,AVM高剂量条件下仍可观察到对脑细胞毒性作用。
Objective To investigate the cytotoxicity of rat avermectins on brain cells by oral administration. Methods The rats were orally administered to observe the occurrence of acute poisoning. The brain tissues of rats were examined by histopathology. The cytotoxicity was determined by MTT assay. Results The symptoms of neurotoxicity were observed after acute poisoning. The changes of degeneration or necrosis of brain cells were observed by histopathological examination. The MTT results showed that the difference was statistically significant (P <0.05) between the 100 mg / kg group and the control group. Conclusion Under the experimental conditions, although the role of the blood-brain barrier, AVM high-dose conditions can still be observed on the cytotoxicity of the brain cells.
