目的对家兔膝关节皮肤进行青藤碱12h体外释放的测定及其释放规律的研究。方法利用体外透皮实验仪模拟体内的环境,以家兔膝关节的皮肤作为渗透媒介,以青藤碱为检测指标,用超高效液相色谱仪测定不同取样点青藤碱的释放量。结果研究表明青藤碱的累计释药量是4.6082(mg·cm-2),累计透过率是58.60%,12h内符合Higuchi方程,方程为Q=1.7357T1/2-1.2501,r值是0.9949。结论青藤碱在家兔膝关节皮肤处12h内释放性良好,家兔膝关节皮肤可用于青藤碱体外释药规律的研究。
OBJECTIVE To study the in vitro release of sinomenine 12 h in rabbit knee joint skin and its release. Methods The in vitro transdermal experimental apparatus was used to simulate the environment in vivo. The rabbit knee joint skin was used as the penetration medium and sinomenine was taken as the detection index. The release of sinomenine from different sampling points was determined by ultra performance liquid chromatography. The results showed that the cumulative release of sinomenine was 4.6082 (mg · cm-2) and the cumulative transmission rate was 58.60%. The Higuchi equation was fitted within 12 hours. The equation was Q = 1.7357T1 / 2-1.2501 and the value of r was 0.9949 . Conclusion Sinomenine released well in the rabbit knee joint skin within 12h. The rabbit knee joint skin could be used to study the in vitro release of sinomenine.
