目的:建立强力五虎合剂中指标性成分苦杏仁苷、盐酸麻黄碱的含量测定方法。方法:应用HPLC检测法(LC-20A岛津高效液相色谱仪),色谱柱为Wonda Sil C18(4.6 mm×250 mm,5μm),流动相分别以甲醇-乙腈-0.1%磷酸(16:4:80),在207 nm波长测定苦杏仁苷;0.1%磷酸-乙腈(95:5),在207 nm波长测定盐酸麻黄碱。结果:苦杏仁苷在0.100~0.800μg(r=0.9994),盐酸麻黄碱在0.008~0.048μg(r=0.9999)之间的范围内呈现良好的线性关系。样品中苦杏仁苷平均回收率为99.51%,RSD=2.51%;盐酸麻黄碱平均回收率为97.47%,RSD=1.75%。结论:该方法结果准确,灵敏度高,重现性好,可用于控制强力五虎合剂的质量。
OBJECTIVE: To establish a method for the determination of amygdalin and ephedrine hydrochloride, an indicator ingredient in Wuhuang Mixture. METHODS: The HPLC method was used for the determination of trace elements in Wonda Sil C18 column (4.6 mm × 250 mm, 5 μm). The mobile phase consisted of methanol-acetonitrile-0.1% phosphoric acid (16: 4 : 80) for the determination of amygdalin at 207 nm and 0.1% phosphoric acid-acetonitrile (95: 5) for determination of ephedrine hydrochloride at a wavelength of 207 nm. Results: Amygdalin showed good linearity in the range of 0.100 ~ 0.800μg (r = 0.9994) and ephedrine hydrochloride in the range of 0.008 ~ 0.048μg (r = 0.9999). The average recoveries of amygdalin in the samples were 99.51% and RSD = 2.51%. The average recoveries of ephedrine hydrochloride were 97.47% and RSD was 1.75%. Conclusion: The method is accurate, sensitive and reproducible. It can be used to control the quality of Wuhuang Mixture.
