目的检测BAT2L蛋白在大鼠脑内的发育学表达情况。方法分别采用Western blot、免疫组化、免疫荧光技术检测BAT2L蛋白在胚胎18d,出生1、7、15d,成年不同时期大鼠脑内的表达、分布、定位情况。结果①Western blot结果显示,BAT2L在胚胎18d,出生1、7d大鼠脑内有高水平表达,而出生15d和成年则表达水平较低。②免疫组化、免疫荧光检测其在大鼠脑内的分布、定位情况:结果显示脑内阳性着色部位为神经元的细胞膜、细胞质,而细胞核、细胞外间质不着色;阳性神经元广泛分布于各个时期的大鼠大脑皮质、小脑皮质、海马、丘脑等部位;阳性神经元的形态在新生阶段和成年大鼠有明显差异,新生大鼠脑组织中部分神经元的突起及分支呈强阳性着色,而成年大鼠神经元突起几乎不着色。结论BAT2L蛋白特异性表达于大鼠大脑的神经元胞体、胞膜及突起中。
Objective To detect the developmental expression of BAT2L protein in rat brain. Methods Western blot, immunohistochemistry and immunofluorescence were used to detect the expression, distribution and localization of BAT2L protein in rat brain at 18d, 1,7d, and 15d respectively. Results ①Western blot results showed that BAT2L was highly expressed in the brain of rats on the 18th day after embryo birth and on the 1st day after birth, while it was lower on the 15th day of birth and the adult. Immunohistochemistry and immunofluorescence were used to detect the distribution and localization in rat brain. The results showed that the positive staining area in the brain was the cell membrane and cytoplasm of neurons, while the nucleus and extracellular matrix were not colored. The positive neurons were widely distributed Cerebral cortices, cerebellar cortex, hippocampus, thalamus and other parts of rats in various periods; positive neuron morphology in neonatal stage and adult rats were significantly different, neonatal rat brain tissue neurons in the process of protuberances and branches were strongly positive Coloring, while neuronal processes in adult rats barely stain. Conclusion BAT2L protein is specifically expressed in the neuronal soma, membrane and protuberances of rat brain.
