以0.06%秋水仙素和2%二甲基亚砜为加倍药剂,比较不同处理时间、不同根长的玉米单倍体幼芽切根处理与直接浸根的加倍效果。结果表明,处理时间12~72 h均能提高玉米单倍体加倍率,显著高于自然加倍,处理48 h加倍率最高。不同根长的切根处理加倍率存在显著差异,根长5~10 mm加倍率最高,根长大于21 mm加倍率最低。切根处理48 h,单倍体植株平均散粉率和结实率分别为51.39%和8.74%,极显著高于直接浸根处理的散粉率和结实率。
With 0.06% colchicine and 2% dimethylsulfoxide as doubling agent, the effects of different times and different root length on the rooting of maize haploid sprout and direct douching rooting were compared. The results showed that the doubling rate of maize haploid could be increased by 12 ~ 72 h, which was significantly higher than that of natural maize. The doubling rate of maize was the highest at 48 h. There were significant differences in rooting rate and root doubling rate between different root lengths. The root length of 5 ~ 10 mm was the highest, the root length was greater than 21 mm, and the least doubling rate was observed. After 48 h of root cutting, the average rate of loose powder and seed setting of haploid plants were 51.39% and 8.74%, respectively, which were significantly higher than those of the direct dip-root treatment.
