用~(60)Coγ射线不同照射量辐照旺盛生长的小麦愈伤组织,并于辐照后3、6、12天取材固定进行电镜观察。结果发现,未经辐照的愈伤组织细胞超微结构正常。经500Rγ射线辐照后,细胞结构有轻度损伤,随之出现旺盛的修复活动,线粒体数量明显增加,内嵴结构完善,内质网发达;辐照后12天细胞结构基本恢复正常。经5000R辐照后细胞严重受损,结构逐趋模糊,细胞器逐步消解,最后细胞死亡。结合辐照后愈伤组织的生长状况以及再生株染色体变异频率等,认为对小麦愈伤组织进行辐射处理时照射量以500~1000R为宜。
The radiated callus was irradiated with ~ (60) Coγ-rays under different irradiation doses. The callus was harvested at 3, 6 and 12 days after irradiation for electron microscopy. The results showed that non-irradiated callus cell ultrastructure is normal. After 500Rγ-ray irradiation, the cell structure was mildly damaged, followed by exuberant repair activities. The number of mitochondria was significantly increased, the internal crest structure was well developed, and the endoplasmic reticulum was developed. The cell structure returned to normal 12 days after irradiation. After 5000R irradiated cells severely damaged, the structure gradually blurred, organelles gradually digestion, the final cell death. Combined with the growth of irradiated callus and the frequency of chromosomal aberrations of regenerative strain, it is considered that the radiation dose of wheat callus is 500 ~ 1000R.
