目的:筛选出血浆中含有抑制血小板聚集的血小板膜糖蛋白GPⅡbⅢa自身抗体的血小板减少性紫癜(ITP)患者,为人源化抗GPⅡbⅢa基因工程抗体的研制做准备。方法:用改良MAIPA法检测24例慢性ITP患者血浆有无血小板膜糖蛋白GPⅡbⅢa自身抗体,用血小板聚集试验研究抗GPⅡbⅢa自身抗体对血小板聚集的影响。结果:24例慢性ITP患者中10例(41.7%)血浆中抗GPⅡbⅢa自身抗体阳性,2例明显抑制血小板聚集功能。结论:少数抗CPⅡbⅢa自身抗体可抑制血小板聚集功能。
Objective: To screen out thrombocytopenic purpura (ITP) patients with platelet glycoprotein GPⅡbⅢa autoantibodies that inhibit platelet aggregation in plasma and prepare for the development of humanized anti-GPⅡb Ⅲ a gene engineering antibody. Methods: The MAIPA method was used to detect the presence or absence of platelet glycoprotein GPⅡb Ⅲ a autoantibodies in 24 patients with chronic ITP. The platelet aggregation test was used to study the effect of anti-GPⅡb Ⅲ a autoantibodies on platelet aggregation. Results: Plasma anti-GPⅡbⅢa autoantibodies were positive in 10 (41.7%) of the 24 patients with chronic ITP and 2 patients were significantly inhibited in platelet aggregation. Conclusion: A few anti-CPⅡbⅢa autoantibodies can inhibit platelet aggregation.
