目的:通过检测兔创伤性下肢深静脉血栓形成P-选择素(CD62P)、血小板活化因子(PAF)及溶酶体膜蛋白(CD63)的表达,探讨葱白提取物防治深静脉血栓的疗效和机制。方法::随机将新西兰白兔24只分为空白模型组、葱白防治组和葱白治疗组。按照赵学凌方法构建兔下肢创伤性深静脉血栓模型,造模成功后,空白模型组给予生理盐水,葱白治疗组给葱白提取物1周,葱白防治组造模前持续给葱白提取物1周,造模后同葱白治疗组。并检测造模前后CD62P、PAF和CD63的变化,对实验数据进行统计分析。结果:造模后,各组兔血清中CD62P、PAF、CD63含量明显升高(P<0.05),葱白防治组上升幅度较小(P<0.05),空白模型组和葱白治疗组上升幅度较大(P<0.01),葱白提取物预防和治疗均可以降低模型组兔血清中CD62P、PAF、CD63含量(P<0.05),和葱白治疗组比,葱白防治组降低CD62P和CD63水平较明显,差异有统计学意义(P<0.05),而对PAF的影响比较差异无统计学意义(P>0.05)。同时葱白提取物预防给药可以降低正常兔血液中CD62P的水平。结论:葱白提取物能防治创伤后深静脉血栓的形成,其机制与减少血小板活化,抗氧化损伤,减少血小板聚集有关。
OBJECTIVE: To investigate the efficacy and mechanism of light blue extract in prevention and treatment of deep vein thrombosis by detecting the expression of P-selectin (CD62P), platelet activating factor (PAF) and lysosomal membrane protein (CD63) . Methods: 24 New Zealand white rabbits were randomly divided into blank model group, light blue control group and light blue treatment group. The model of traumatic deep vein thrombosis of lower extremities was constructed according to Zhao Xueling’s method. After the model was established successfully, the model group was given normal saline, the light blue treatment group was given the light blue extract for 1 week, the light blue prevention group was given the light blue extract for 1 week before modeling After the same light blue treatment group. The changes of CD62P, PAF and CD63 before and after the model were detected, and the experimental data were statistically analyzed. Results: After modeling, the levels of CD62P, PAF and CD63 in serum of rabbits in each group were significantly increased (P <0.05), those in control group were lower (P <0.05), those in blank model group and light blue treatment group were higher (P <0.01). Preventive and curative effects of extract of scutellaria barbata could reduce the levels of CD62P, PAF and CD63 in the model group (P <0.05). Compared with the scutellar group, the levels of CD62P and CD63 decreased significantly There was statistical significance (P <0.05), but there was no significant difference in the effect on PAF (P> 0.05). In the meantime, prophylactic administration of scutellaria extract can reduce the level of CD62P in normal rabbit blood. Conclusion: Scallion extract can prevent and treat post-traumatic deep vein thrombosis. Its mechanism is related to reducing platelet activation, anti-oxidative damage and reducing platelet aggregation.
