为了观察大鼠脑缺血再灌注损伤后大脑皮质TNF-α及其mRNA表达的时程变化,本实验采用大脑中动脉线栓法制作大鼠局灶性脑缺血再灌注模型,用免疫组化染色法检测不同再灌注时间大脑皮质TNF-α的蛋白表达水平,以及用逆转录-多聚酶链反应法(RT-PCR)检测脑皮质内TNF-αmRNA的表达情况。结果显示:脑缺血1h,再灌注12h后大脑皮质内TNF-α及其mR-NA的表达开始升高,24h后达到高峰,随后逐渐下降。两组大脑皮质TNF-α及其mRNA的表达水平差异有统计学意义(P<0.01),模型组的TNF-α及其mRNA的表达水平明显高于假手术组。本研究结果表明,TNF-α在大鼠脑缺血再灌注损伤的调节过程中发挥重要作用。
In order to observe the time course of TNF-α and its mRNA expression in cerebral cortex after cerebral ischemia-reperfusion injury in rats, a rat model of focal cerebral ischemia-reperfusion was established by middle cerebral artery occlusion method. The protein expression of TNF-α in cerebral cortex at different reperfusion time and the expression of TNF-αmRNA in cerebral cortex were detected by reverse transcription-polymerase chain reaction (RT-PCR). The results showed that the expression of TNF-α and mR-NA began to increase in cerebral cortex 1h after cerebral ischemia and 12h after reperfusion, peaked at 24h and then decreased gradually. The expression of TNF-α and its mRNA in the cerebral cortex of the two groups were significantly different (P <0.01). The expression of TNF-α and its mRNA in the model group was significantly higher than that in the sham-operated group. The results of this study indicate that TNF-α plays an important role in the regulation of cerebral ischemia-reperfusion injury in rats.
