目的探讨磷脂酰乙醇胺结合蛋白4(PEBP4)基因对于非小细胞肺癌(NSCLC)细胞体外生物学行为的影响。方法体外培养NSCLC细胞系HCC827细胞,并分为A组(PEBP4shRNA转染)、B组(无关序列转染)和C组(不做任何处理)。采用Western blot法检测PEBP4、周期蛋白D1、p53、Bcl-2、MMP-2和MMP-9蛋白的表达,MTT法、流式细胞仪和Transwell实验分析PEBP4对NSCLC细胞生物学行为的影响。结果PEBP4基因在NSCLC细胞系中呈高表达。与B、C组比较,A组细胞的活力及迁移能力降低,细胞凋亡增加,周期蛋白D1、Bcl-2、MMP-2及MMP-9蛋白表达水平降低,p53蛋白表达增加(P<0.05)。结论 PEBP4基因可促进HCC827细胞活力和迁移能力,抑制细胞凋亡。
Objective To investigate the effect of phosphatidylethanolamine binding protein 4 (PEBP4) gene on the biological behavior of non-small cell lung cancer (NSCLC) in vitro. Methods NSCLC cell line HCC827 cells were cultured in vitro and divided into group A (PEBP4 shRNA transfection), group B (unrelated sequence transfection) and group C (without any treatment). Western blot was used to detect the expression of PEBP4, Cyclin D1, p53, Bcl-2, MMP-2 and MMP-9. MTT assay, flow cytometry and Transwell assay were used to analyze the effect of PEBP4 on the biological behavior of NSCLC cells. Results PEBP4 gene was highly expressed in NSCLC cell lines. Compared with group B and group C, the viability and migration ability of group A were decreased, the apoptosis of group A was increased, the expression of cyclin D1, Bcl-2, MMP-2 and MMP-9 were decreased and the expression of p53 was increased ). Conclusion PEBP4 can promote the viability and migration of HCC827 cells and inhibit the apoptosis of HCC827 cells.
