[目的]改进聚丙烯酰胺凝胶电泳检测致病疫霉菌SSR标记方法。[方法]以2009年在宁夏固原地区采集的马铃薯晚疫病样为试验材料,提取其DNA,PCR扩增,并将其产物进行聚丙烯酰胺凝胶电泳。[结果]采用12%聚丙烯凝胶电泳检测引物D13、G11、PI02,8%聚丙烯凝胶电泳检测引物PI4B、PI63、SSR4、SSR8、SSR11,最后采用0.1%硝酸银染色,取得了比较理想的电泳及染色效果。[结论]该研究建立的适用于致病疫霉菌SSR标记的电泳和染色技术方法具有检测灵敏度高、操作简单、获得的条带清晰等特点,是一种行之有效、简便快捷的检测方法。
[Objective] The research aimed to improve the SSR labeling method of Phytophthora infestans by polyacrylamide gel electrophoresis. [Method] With potato late blight samples collected in 2009 in Ningxia Guyuan as experimental materials, the DNAs were extracted and amplified by PCR. The products were subjected to polyacrylamide gel electrophoresis. [Result] The primers PI3B, PI63, SSR4, SSR8 and SSR11 were detected by 12% polypropylene gel electrophoresis (D13, G11, PI02, 8% polypropylene gel electrophoresis) The electrophoresis and staining results. [Conclusion] The method of electrophoresis and staining for SSR markers of Phytophthora infestans was established in this study. It has high detection sensitivity, simple operation and clear bands. It is an effective, simple and rapid detection method.
