目的应用常规染色体分析研究青蒿素对p53功能突变的人乳腺癌细胞MDA-MB-435的放射敏感作用。方法通过MTT法测定不同药物浓度及不同药物作用时间对细胞毒性的影响,常规染色体制片,计数细胞分类项中双着丝粒体(dic)和着丝粒环(r)数量,将经过青蒿素作用的乳腺癌细胞染色体畸变率(dic+r)同单纯照射的细胞染色体畸变率进行统计分析。结果青蒿素对实验细胞MDA-MB-435毒性较小,200μmol/L浓度时作用24h细胞存活率>95%,48h细胞存活率>80%。参考存活曲线确定常规染色体畸变分析实验的药物浓度为200μmol/L,作用时间为24h。将得到的数据进行回归拟合,可以得到药物作用及未经药物作用的剂量效应曲线,比较两组曲线,青蒿素作用组的染色体畸变率明显高于单纯照射细胞(P>0.05)。结论青蒿素能增加辐射对乳腺癌细胞MDA-MB-435的染色体损伤。
Objective To study the radiosensitization effect of artemisinin on human breast cancer cell line MDA-MB-435 with p53 mutation by routine chromosome analysis. Methods MTT assay was used to determine the effect of different drug concentrations and different drug action time on cytotoxicity. The number of dicentric and centromere (r) The chromosome aberration rate (dic + r) of breast cancer cells treated with artemisinin was statistically analyzed with the chromosomal aberration rate of cells exposed to light. Results Artemisinin had less cytotoxic effect on MDA-MB-435 cells. The cell viability was> 95% at 200μmol / L for 24 hours and> 80% after 48 hours. Conclusion Artemisinin can increase the chromosome damage of breast cancer cell line MDA-MB-435.
