目的 :建立重组人白介素 - 11的高效凝胶色谱含量测定方法。方法 :采用外标法 ,色谱柱 :BiosepSEC -S2 0 0 0 (30 0mm× 7 8mm) ,流动相 :0 2mol·L-1磷酸盐缓冲液 (pH 6 8) ,流速 :0 5mL·min-1,检测波长 :2 80nm。结果 :线性范围 :2 5～ 30 μg (r =0 9990 ) ,平均回收率 10 1 1%～ 10 1 9% (RSD<1 8% ,n =9)。精密度 :RSD=0 75 % (n=5 ) ;最低检测限 0 15 μg。 结论 :该法用于测定重组人白介素 - 11的含量 ,简便 ,准确 ,重现性好。
Objective: To establish a method for the determination of recombinant human interleukin - 11 by high performance gel permeation chromatography. Methods: The external standard method was used. The chromatographic column was Biosep SEC-200 (30 0 mm × 78 mm). The mobile phase was 0 2 mol·L-1 phosphate buffer (pH 6 8) 1, detection wavelength: 2 80nm. RESULTS: The linear range was 25-30 μg (r = 0 9990) with an average recovery of 101.1% -101.9% (RSD <18%, n = 9). Precision: RSD = 0 75% (n = 5); the lowest limit of detection was 0 15 μg. Conclusion: The method was used to determine the content of recombinant human interleukin - 11, which was simple, accurate and reproducible.
