目的:研究二七提取液衍生化(本文采用了乙酰化)和蒸发光散射检测器(evaporative light-scatteringdetector,ELSD)在中药三七组分分析和鉴定中的应用。方法:采用乙腈-水梯度洗脱、紫外检测的方法,比较乙酰化前后的三七样品提取液的高效液相色谱(high pedbrmance liqmd chromatography,HPLC)图谱差异;分别采用乙腈-水(体积比30∶70)和乙腈-水(体积比85∶15)作为流动相,比较用 ELSD 和紫外检测器时三七样品提取液的HPLC 图谱差异。结果:在三七提取液乙酰化后,可以观察到在原样品色谱图中较早洗脱出的组分的变化,其中某些组分衍生物的保留时间大大延长。分别用 ELSD 检测和紫外检测可以得到不同的色谱图,用 ELSD 检测方法可以得到几种不同变化方式的峰群,从而提供了更多的三七提取液化学组成的信息。结论:三七提取液乙酰化后,其中一些亲水组分的极性大大降低,那些在反相色谱中较早流出、较难分离的组分经衍生化后出峰状况得到改善,ELSD 还可以检测到有药理作用但因没有紫外吸收较难检测的糖类,这些都给中药三七的分析和鉴定提供了新的思路
Objective: To study the application of the derivatization of Erqi extract (acetylation in this article) and evaporative light-scattering detector (ELSD) in the analysis and identification of Panax pseudoginseng. METHODS: Acetonitrile-water gradient elution and UV detection methods were used to compare the differences of high pedbrmance liqmd chromatography (HPLC) profiles of the extracts of P. notoginseng before and after acetylation; acetonitrile-water (volume ratio 30) was used respectively. : 70) and acetonitrile-water (volume ratio 85:15) as mobile phase, comparing HPLC chromatogram differences of the Panax notoginseng sample extracts with ELSD and UV detectors. RESULTS: After acetylation of Panax pseudoginseng extract, changes in the components eluted earlier in the chromatogram of the original sample were observed, and the retention time of some of the component derivatives was greatly prolonged. Different chromatograms can be obtained with ELSD detection and UV detection, respectively, and ELSD detection methods can be used to obtain peaks with several different variations, thus providing more information on the chemical composition of Panax notoginseng extracts. Conclusion: After the acetylation of Panax pseudoginseng extract, the polarity of some of its hydrophilic components is greatly reduced, and the out-of-peak condition is improved after the derivatization of components that elute earlier in reversed-phase chromatography. Can detect pharmacological effects but there is no more difficult to detect the absorption of sugar in the UV, these all provide a new idea for the analysis and identification of traditional Chinese medicine Sanqi
