目的研究南海深海沉积物来源真菌Penicillium sp.F00120的次级代谢产物及抗菌活性。方法采用硅胶、Sephadex LH-20、制备薄层进行分离纯化,通过波谱分析(1H-NMR、13C-NMR)鉴定其结构;采用滤纸片琼脂扩散法测试化合物的抗菌活性。结果从其发酵物中分离得到8个化合物,分别鉴定为麦角甾-4,6,8(14),22-四烯-3-酮(1),25-羟基-麦角甾-4,6,8(14),22-四烯-3-酮(2),麦角甾-7,22-二烯-3β,5α,6β-三醇(3),麦角甾-5,7,22-三烯-3β-醇(4),5α,8α-环二氧-6,22-麦角甾二烯-3β-醇(5),5α,6α环氧麦角甾-8(14),22-二烯-3β,7α-二醇(6),胆甾醇(7)和对羟基苯乙酮(8)。结论化合物1~3、6~8为首次从该菌中分离得到;化合物3、4和8具有一定的抗菌活性。
Objective To study the secondary metabolites and antibacterial activity of fungi Penicillium sp. F00120 from deep-sea sediment in the South China Sea. Methods Silica gel and Sephadex LH-20 were used for the isolation and purification. The structures of the compounds were identified by spectral analysis (1H-NMR, 13C-NMR). The antibacterial activity of the compounds was tested by filter paper agar diffusion method. As a result, eight compounds were isolated from the fermented products thereof and identified as ergosta-4,6,8 (14), 22-tetraen-3-one (1), 25-hydroxy- (2), ergosta-7,22-diene-3β, 5α, 6β-triol (3), ergistetra-5,7,22-triene (5), 5α, 6α-epoxygalaserin-8 (14), 22-diene-3β- 3β, 7α-diol (6), cholesterol (7) and p-hydroxyacetophenone (8). Conclusion Compounds 1 ~ 3, 6 ~ 8 are isolated from this strain for the first time. Compounds 3, 4 and 8 have some antibacterial activity.
