目的 探讨MDR1 基因对骨髓细胞的抗癌药物毒性防护作用,寻找防治抗癌药物所致骨髓抑制的有效措施.方法 以小鼠骨髓细胞为靶细胞,通过脂质体介导,采用MDR1 表达质粒PHAMDR1/A,将MDR1 基因转入骨髓细胞.分别采用流式细胞术、免疫组化和罗丹明试验观察MDR1 基因的表达及表达产物的功能,并通过集落细胞培养和抗性试验及骨髓移植动物模型分别从体外、体内观察骨髓细胞对抗癌药物的抗性.结果 成功地将MDR1 基因导入了小鼠骨髓细胞,并在体内外均获得了表达、对抗癌药产生了明显的抗性.结论 MDR1 基因的骨髓细胞转导可保护骨髓细胞免受抗癌药物的损伤.“,”Objective To investigate the protective effects of mdr1 gene on hematopoietic cells against anticancer agents and to explore an effective approach to prevent myelosuppression due to chemotherapy Methods mdr1 gene was transferred into murine bone marrow cells with plasmid pHaMDR1/A by the mediation of lipofectin Pgp ,a product of mdr1 gene expression , and its function were detected by flow cytometry, immunohistochemistry,and rhodamine test respectively Also, the resistance of transferred murine bone marrow cells to anticancer agents was tested by colony forming unit test after exposure to anticancer agents and by bone marrow transplantation murine model Results mdr1 gene was successfully transferred and expressed in bone marrow cells Transferred murine bone marrow cells were resistant to anticancer agents both in vitro and in vivo Conclusions Transfection of mdr1 gene into hematopietic cells can protect bone marrow cells against the toxicities of anticancer agents
