本文采用荧光偏振免疫(FPIA)法测定环孢素A(CsA)的全血浓度。测定对象为肾移植患者37例,其中5例患者口服CsA后进行药代动力学研究,按口服吸收二房室模型计算药代动力学参数。结果表明患者之间?着显著的个体差异。同时我们还对37例肾移植患者进行169次临床血药浓度监测。结果也表明患者个体之间差异较大。对制定给药方案、肾毒性和排斥反应的鉴别,以及药物相互作用影响,正确的服药方法等都有重要的临床意义。
In this paper, the whole blood concentration of cyclosporin A (CsA) was measured by fluorescence polarization immunoassay (FPIA). Totally, 37 patients with renal allograft were enrolled in this study. Five of them received pharmacokinetic study after oral administration of CsA. Pharmacokinetic parameters were calculated by oral absorption of two-compartment model. The results showed significant individual differences between patients. At the same time, we also carried out 169 clinical plasma concentrations monitoring on 37 renal transplant patients. The results also show that patients vary widely among individuals. To develop dosing regimens, identification of nephrotoxicity and rejection, as well as the impact of drug interactions, the right medication and so on have important clinical significance.
