为了确定DM-2型大孔树脂分离、纯化沙枣多糖的最佳工艺条件,试验以吸附率和解吸率为评价指标,通过动态试验研究各因素对分离、纯化效果的影响。结果表明:在沙枣多糖样品溶液浓度为0.7 mg/mL、pH值为10、上样速率为2.5 BV/h、上样量为3 BV、洗脱剂浓度为55%、洗脱速率为1 BV/h、洗脱剂用量为3 BV时,吸附率和解吸率分别达到84.05%和97.3%。说明在同时满足上述条件的情况下,DM-2型大孔树脂分离、纯化沙枣多糖的效率较高。
In order to determine the optimal conditions for the separation and purification of polysaccharide from P. emarginata, the adsorption and desorption rates of DM-2 macroporous resin were used as evaluation indexes to study the influence of various factors on the separation and purification effect through dynamic test. The results showed that when the sample concentration was 0.7 mg / mL, the pH value was 10, the sample loading rate was 2.5 BV / h, the sample volume was 3 BV, the eluent concentration was 55% and the elution rate was 1 BV / h, the amount of eluent 3 BV, the adsorption rate and desorption rate reached 84.05% and 97.3%. It shows that DM-2 macroporous resin can purify P. purpurea polysaccharide efficiently while satisfying the above conditions.
