为研究肝素对胸腺细胞发育的影响,以FTOC的方法进行体外器官培养,以不同浓度肝素干预其发育。观察发育不同时间点,胸腺细胞数和胸腺细胞表型的变化情况。结果显示:在4U/ml的肝素浓度下,胸腺细胞数比对照组显著减少,尤其是CD4+CD8+双阳性细胞数变化明显,且胸腺细胞数的减少与肝素用量具有明显剂量依赖关系;另外,在体外培养的第3天,肝素干预组中胸腺细胞37.98%表达CD69分子,显著高于对照组的0.76%。这些证据提示肝素能够影响双阳性胸腺细胞的发育。
In order to study the effect of heparin on the development of thymocytes, the in vitro organ culture was performed by the method of FTOC, and its development was interfered with different concentrations of heparin. The changes of thymus and thymocyte phenotypes at different time points were observed. The results showed that under the condition of 4U / ml heparin concentration, the number of thymocytes was significantly decreased, especially the number of double positive cells of CD4 + CD8 +, and the decrease of thymocyte number was significantly dose dependent. In addition, On day 3 of in vitro culture, 37.98% of the thymocytes in the heparin-treated group expressed CD69 molecules, which was significantly higher than that of the control group (0.76%). These evidence suggest that heparin can affect the development of double positive thymocytes.
