目的:探索连翘花水提物(FFA)是否对自由基所致的线粒体损伤有保护作用,并研究其抗氧化能力、超氧阴离子的清除活性及抗衰老机制。方法:采用硫代巴比妥酸(TBA)比色法测定丙二醛(MDA)含量、分光光度法测线粒体的肿胀度,并以还原型辅酶I-氮蓝四唑-吩嗪硫酸甲酯(NADH-NBT-PMS)为超氧阴离子生成系统测定对超氧阴离子的清除能力。小鼠颈背部皮下注射D-半乳糖(D-gal)100 mg/kg.d,连续给药7周造衰老模型,利用钼酸胺比色法、黄嘌呤氧化酶法、二硫代二硝基苯甲酸比色法、Fenton反应显色法分别测定FFA对小鼠肝匀浆过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)及抗羟自由基活力的影响。结果:FFA可明显抑制脑、肝匀浆及肝线粒体的脂质过氧化、线粒体肿胀,并呈剂量依赖关系,清除超氧阴离子的能力显著,且能明显增强CAT、SOD、GSH-Px及抗羟自由基活力。结论:FFA能通过清除活性氧自由基及提高抗氧化酶活力来保护线粒体,具有一定的抗氧化及抗衰老药用价值。
OBJECTIVE: To explore whether FFA of Forsythia suspensa has a protective effect on free radical-induced mitochondrial damage and its anti-oxidative capacity, superoxide anion scavenging activity and anti-aging mechanism. Methods: The content of malondialdehyde (MDA) was determined by thiobarbituric acid (TBA) colorimetric method. The degree of swelling of mitochondria was measured by spectrophotometry. The content of reduced coenzyme I-TNT-phenazine methosulfate (NADH-NBT-PMS) is a superoxide anion generating system for the determination of superoxide anion scavenging ability. Mice were subcutaneously injected with D-galactose (100 mg / kg.d) on the back of the neck for 7 weeks. The mice were sacrificed by the method of molybdate amine colorimetry, xanthine oxidase method, Benzoic acid colorimetry and Fenton reaction chromogenic assay were used to determine the effects of FFA on liver homogenate catalase (CAT), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) And anti-hydroxyl radical activity. Results: FFA significantly inhibited lipid peroxidation and mitochondria swelling in brain, liver homogenate and liver mitochondria, and showed a dose-dependent relationship with the ability of scavenging superoxide anion and significantly enhancing CAT, SOD, GSH-Px and anti- Hydroxyl radical activity. Conclusion: FFA can protect mitochondria by removing reactive oxygen species and increasing antioxidant enzyme activity, which has certain anti-oxidative and anti-aging medicinal value.
