目的建立高效液相色谱法(HPLC)直接测定各类食品中吊白块的特异性强,准确度高的方法。方法食品中的吊白块遇碱分解成甲醛和亚硫酸盐,在酸性条件下与盐酸副玫瑰苯胺生成紫红色产物,该产物在HPLC上用离子对试剂与乙腈做流动相,通过C18柱分离,利用紫外检测器(或二极管阵列检测器),在554nm波长下测定。结果其产物有很高的响应值,其色谱峰面积与吊白块的含量呈正相关,r=0.9995,线性范围为0.02~0.6mg,回收率93.5%~102.2%,检出限0.003mg,RSD=9.8%。结论该方法特异性强,准确度高,重复性好,线性范围宽,适用于对各类食品中添加吊白块的检测。
OBJECTIVE To establish a HPLC-based method for the direct determination of strychnos lites in various foods with high specificity and high accuracy. Methods When the hanging white lumps in food were decomposed into formaldehyde and sulfite under alkaline condition, the purple rose product was formed with pararosaniline hydrochloride under acidic condition. The product was separated on a C18 column using ion pair reagent and acetonitrile as the mobile phase on HPLC , Using UV detector (or diode array detector), measured at a wavelength of 554nm. The results showed that the product had a high response value, and the peak area of the product was positively correlated with the content of hydrolysates. The linear range was 0.02 ~ 0.6mg, the recovery was 93.5% ~ 102.2%, the detection limit was 0.003mg, RSD = 9.8%. Conclusion The method is highly specific, accurate, reproducible and has a wide linear range. It is suitable for the detection of various kinds of foodstuffs.
