目的:开郁胶囊是本室自行研制的由8种传统中药组成的中药新复方制剂,本研究观察开郁胶囊对数种抑郁动物模型的影响。方法:采用ICR小鼠随机分为对照组、开郁胶囊低、中、高剂量组(按生药量计为6.6,13.2,26.4 g.kg-1)及阳性对照药帕罗西汀组。连续ig 14 d,于末次给药1.5 h后采用小鼠悬尾、强迫游泳、5-羟色胺酸(5-HTP)诱导甩头及利血平拮抗实验动物模型,评价开郁胶囊的抗抑郁作用。结果:与对照组相比,开郁胶囊中、高剂量组能显著缩短小鼠悬尾不动时间(P<0.05);开郁胶囊高剂量组显著增加5-HTP诱导的小鼠甩头次数(P<0.01),缩短甩头潜伏期(P<0.05)。注射利血平后动物出现利血平化,即眼睑下垂、体温下降及活动抑制;开郁胶囊中、高剂量ig 14 d,可改善利血平化小鼠的眼睑下垂及活动抑制(P<0.01),对体温下降亦有显著的拮抗作用(P<0.01)。结论:开郁胶囊对拟抑郁症小鼠模型具有明显的拮抗作用,其作用机制可能与增强5-羟色胺能和去甲肾上腺素能神经功能有关。
OBJECTIVE: Kaiyu capsule is a new compound preparation of Chinese traditional medicine consisting of 8 kinds of traditional Chinese medicines developed in our laboratory. This study observed the influence of Kaiyu capsule on several kinds of depression animal models. Methods: ICR mice were randomly divided into control group, Kaiyu capsule low, medium and high dose groups (6.6,13.2,26.4 g.kg-1 according to the crude drug amount) and paroxetine positive control group. After 14 days of continuous administration, mice were subjected to tail suspension, forced swimming, 5-HT-induced head-shark and reserpine antagonistic animal models for 1.5 h after the last administration, to evaluate the antidepressant effect of Kaiyu capsule . Results: Compared with the control group, Kaoyu capsule medium and high dose groups can significantly reduce the tail suspension time (P <0.05); Kaoyu capsule high dose group significantly increased 5-HTP-induced mouse shampooing times (P <0.01), shortening the head-sham incubation period (P <0.05). After injection of reserpine, the animals appeared reserpine, that is, eyelid ptosis, hypothermia and activity inhibition; Kaiyu capsule, high dose ig 14 d, can improve reserpine-induced mice’s ptosis and activity inhibition (P < 0.01), but also had a significant antagonism on the decrease of body temperature (P <0.01). Conclusion: Kaiyu capsule has a significant antagonistic effect on mice model of depression, and its mechanism may be related to the enhancement of the neurotransmission of serotonergic and noradrenergic nerves.
