目的 探讨不同浓度的单核细胞趋化因子 (MCP 1)对肿瘤浸润淋巴细胞 (TIL)增殖的作用 ,以及能否增加TIL对膀胱肿瘤细胞的细胞毒性。方法 观察在不同浓度的MCP 1(0 g·L-1,1× 10 -4g·L-1,1× 10 -3 g·L-1,1× 10 -2 g·L-1)作用下 ,不同时间相对应的TIL增殖的变化。用MTT法测定不同浓度的MCP 1作用后TIL对细胞毒性的影响。结果 MCP 1(1× 10 -3 g·L-1)作用 2周后 ,可显著增高TIL增殖 (P <0 .0 1) ,加强细胞杀伤率 (16 .4 % ) (P <0 .0 1)。结论 一定浓度的MCP 1可促进TIL细胞的增殖 ,并可增强TIL对EJ细胞的细胞毒性。
Objective To investigate the effect of different concentrations of monocyte chemoattractant (MCP 1) on the proliferation of tumor infiltrating lymphocytes (TILs) and the cytotoxicity of TIL on bladder tumor cells. Methods The effects of different concentrations of MCP 1 (0 g · L -1, 1 × 10 -4 g · L -1, 1 × 10 -3 g · L -1,1 × 10 -2 g · L -1) , Different time corresponding TIL proliferation changes. The effect of TIL on cytotoxicity was assayed by MTT assay with different concentrations of MCP1. Results After treated with MCP 1 (1 × 10 -3 g · L -1) for 2 weeks, the proliferation of TIL was significantly increased (P <0.01) and the cell killing rate was enhanced (16.4%) (P <0. 0 1). Conclusion A certain concentration of MCP 1 can promote the proliferation of TIL cells and enhance the cytotoxicity of TIL on EJ cells.
