目的基于UPLC-代谢组学技术研究茯苓皮结合有氧运动对高脂血症大鼠的影响。方法雄性SD大鼠,分为5组,每组8只。高脂饲料喂饲大鼠4周,同时进行4周的茯苓皮、有氧运动、茯苓皮结合有氧运动治疗。实验第1天和第2周末尾静脉取血,第4周后处死动物,采用UPLC-代谢组学方法推测高脂血症模型是否成功。比较血清TG、TC、HDL-C及LDL-C含量,同时测定血清CAT、GSH-Px、MDA和SOD的浓度。结果在4周时间,代谢组学和生化指标证明高脂血模型成功建立,不同时间段产生不同的损伤程度且明确了高脂血症模型的时间进展轨迹。生化指标显示茯苓皮和有氧运动均可显著降低大鼠血清TG、TC和LDL-C提高、HDL-C水平;茯苓皮结合有氧运动能显著升高大鼠血清GSH-Px、SOD、CAT水平,同时降低MDA的水平,脂质过氧化产物水平显著降低。结论(1)代谢组学结果显示高脂血症模型已成功建立;(2)茯苓皮结合有氧运动可治疗高脂血症大鼠血脂异常,并有较明显的抗脂质过氧化作用;(3)代谢组学可以作为有氧运动治疗疾病程度及效果的一个新的判断方法。
OBJECTIVE: To study the effect of Poria Cortex combined with aerobic exercise on hyperlipidemic rats based on UPLC-metabolomics. Methods Male Sprague-Dawley rats were divided into 5 groups with 8 rats in each group. High-fat diet for 4 weeks, while 4 weeks of Poria skin, aerobic exercise, Poria skin combined with aerobic exercise therapy. Animals were sacrificed on the tail vein on the first day and the second week of the experiment, and animals were sacrificed after the fourth week. The hyperlipidemia model was estimated by UPLC-metabolomics. Serum levels of TG, TC, HDL-C and LDL-C were measured and serum concentrations of CAT, GSH-Px, MDA and SOD were measured simultaneously. Results At 4 weeks, the hyperlipemia model was established successfully by metabolomics and biochemical indexes. Different injury stages were observed at different time points and the path of hyperlipemia was identified. Biochemical indicators showed that both Poria cocos and aerobic exercise can significantly reduce serum TG, TC and LDL-C levels and HDL-C levels in rats; Poria cortex combined with aerobic exercise can significantly increase serum GSH-Px, SOD, CAT levels , While reducing the level of MDA, lipid peroxidation product levels significantly reduced. Conclusions (1) Metabolomics results show that hyperlipidemia model has been successfully established; (2) Poria cortex combined with aerobic exercise can treat dyslipidemia and hyperlipidemia in hyperlipidemic rats; (3) Metabolomics can be used as a new method of judgment for the degree and effect of aerobic exercise in treating diseases.
