利用邻-甲酚酞络合剂为显色剂建立了连续测定血清钙和镁的吸光光度法。钙和镁与该显色剂在pH11的碱性缓冲液中形成紫色络合物,于570nm波长处均有最大吸收,且曲线具有加和性。先测得钙镁吸光度之和,加入8-羟基喹啉以掩蔽镁离子的干扰后测得钙的吸光度,两者之差即为镁的吸光度。钙和镁的摩尔吸光系数分别为2.14×10~4和1.51×l0~4。方法应用于血清样品中钙和镁的连续测定,结果令人满意。
A spectrophotometric method for the continuous determination of serum calcium and magnesium was established by using o - cresolphthalein complexing reagent as chromogenic reagent. Calcium and magnesium and the developer form a purple complex in an alkaline buffer at pH 11 with maximum absorption at a wavelength of 570 nm and the curves are additive. First measured the sum of calcium and magnesium absorbance, adding 8-hydroxyquinoline to mask the interference of magnesium ions measured absorbance of calcium, the difference between the two is the absorbance of magnesium. The molar absorptivities of calcium and magnesium were 2.14 × 10 ~ 4 and 1.51 × 10 ~ 4, respectively. The method was applied to the continuous determination of calcium and magnesium in serum samples with satisfactory results.
