应用细胞融合技术将小鼠MHC基因引入人胃癌细胞,建立了1株杂交瘤细胞系。经染色体分析、流式细胞仪及ABC免疫组化染色证实,其具有杂交瘤细胞的特征,可同时表达人胃癌抗原和小鼠MHC分子。免疫学特性研究显示,该杂交瘤细胞能被小鼠肿瘤特异性杀伤细胞所识别和杀灭,有可能成为人胃癌疫苗研究模型中较理想的靶细胞。本项研究结果的意义是,在应用动物模型研究人类肿瘤抗原及疫苗时,进行种间细胞融合可能是克服靶细胞与效应细胞之间MHC不匹配的简单而有效的方法。
Cell fusion technique was used to introduce mouse MHC gene into human gastric cancer cells and a hybridoma cell line was established. Chromosomal analysis, flow cytometry, and ABC immunohistochemical staining confirmed that the hybridoma cells possess the characteristics of hybridoma cells and can simultaneously express human gastric cancer antigens and mouse MHC molecules. Studies of immunological properties have shown that the hybridoma cells can be recognized and killed by mouse tumor-specific killer cells, and may become ideal target cells in a human gastric cancer vaccine research model. The significance of the results of this study is that when animal models are used to study human tumor antigens and vaccines, inter-species cell fusion may be a simple and effective method to overcome MHC mismatch between target cells and effector cells.
