目的:探讨四物合剂提取纯化过程中不同中间产物补血及调经止痛的功效,以期为其生产工艺研究提供科学、合理的药效学评价基础。方法:按照现行版中国药典收载四物合剂项下制备当归-川芎蒸馏液、四物水煎液、四物醇沉上清液、四物醇沉淀样品液,采用乙酰苯肼(APH)-环磷酰胺(CTX)联用构建大鼠血虚模型,观察对其外周血象、脏器指数的影响;采用雌二醇-缩宫素联用构建小鼠痛经模型,观察其对血浆前列腺素2α(PGF_(2α))、前列腺素E2(PGE_2)及子宫组织丙二醛(MDA)、钙离子(Ca~(2+))的影响。结果:当归-川芎蒸馏液、四物水煎液、四物醇沉上清液、四物醇沉淀样品液均能显著升高血虚模型大鼠白细胞(WBC)、红细胞(RBC)、血红蛋白(HGB)、红细胞比容(HCT)及肝脏、胸腺指数,四物醇沉上清液能提高血虚大鼠PLT水平,当归-川芎蒸馏液、四物水煎液能显著升高血虚大鼠脾脏指数。当归-川芎蒸馏液、四物水煎液、四物醇沉上清液、四物醇沉淀样品液均能显著升高痛经模型小鼠血浆PGE_2,降低血浆PGF_(2α)及子宫组织MDA含量;当归-川芎蒸馏液、四物水煎液、四物醇沉上清液均可显著降低痛经模型小鼠子宫组织Ca~(2+)含量。结论:四物合剂提取纯化工艺能较好的保留方中药效成分,发挥补血、调经止痛功效;同时发现被醇沉除去的物质仍具有一定的药效。
OBJECTIVE: To explore the efficacy of blood transfusions and menstruation and analgesia of different intermediates during the process of extraction and purification of Siwu Mixture, in order to provide a scientific and rational pharmacodynamic evaluation basis for the study of its production technology. Methods: According to the Chinese version of Chinese Pharmacopoeia containing Siwu Mixture, Angelica sinensis - Chuanxiong Distillate, Siwu Decoction, the four alcohol precipitation supernatant, the four alcohol precipitation sample solution were prepared, and acetophenhydrazine (APH) CTX was used to establish the model of blood deficiency in rats to observe its effect on peripheral blood and viscera index. Estradiol and oxytocin were used to establish mice model of dysmenorrhea, and its effects on plasma prostaglandin 2α (PGF 2α), prostaglandin E2 (PGE 2) and uterine malondialdehyde (MDA) and calcium (Ca 2+) were measured. Results: Angelicae sinensis extract, Chuanxiong decoction, Siwu decoction, four alcohol precipitation supernatant and four alcohol precipitation samples could significantly increase WBC, RBC and hemoglobin HGB), hematocrit (HCT) and liver, thymus index, four alcohol precipitation supernatant can increase PLT levels in blood deficiency rats, Angelica - Chuanxiong Distillate, Siwu decoction can significantly increase blood deficiency rats Spleen index. Angelicae sinensis extract, Chuanxiong decoction, Siwu Decoction, Siwu alcohol Shen supernatant and Siwu alcohol precipitation samples can significantly increase plasma PGE_2, decrease plasma PGF 2α and uterine tissue MDA content in dysmenorrheal model mice. Angelica - Chuanxiong Distillation, Siwu Decoction, four alcohol Shen supernatant can significantly reduce the dysmenorrhea model mice uterine tissue Ca ~ (2+) content. Conclusion: The extraction and purification process of Siwu Mixture can better retain the pharmacodynamics of prescription and exert the blood-activating and analgesic-analgesic effects. At the same time, it is found that the substances removed by alcohol precipitation still have certain pharmacodynamics.
