目的:研究肠胃清方对大肠癌耐药细胞株HCT-8/V的逆转作用及其抑制P-gp表达的影响。方法:MTT法测定肠胃清对HCT-8/V细胞株的逆转作用,流式细胞术测定P糖蛋白(P-gp)的表达。结果:HCT-8/V细胞株对VCR、DDP、5-FU、MMC的耐药指数分别为18.6、22.3、4.3、3.7。经肠胃清方药物血清作用48h后,其对化疗的耐药性显著下降,对VCR、DDP的逆转指数分别为3.3和7.5,静息状态下HCT-8/V细胞的P-gp表达显著高于HCT-8(P<0.01),而经肠胃清药物血清作用后P-gp表达显著降低(P<0.01)。结论:肠胃清方可以逆转人大肠癌耐药细胞株HCT-8/V的多药耐药性,其机制可能与其下调P-gp的表达有关。
Objective: To study the effect of “Wei Wei Qing Fang” on the reversal of multidrug resistance in human colorectal cancer cell line HCT-8 / V and its effect on the expression of P-gp. Methods: MTT assay was used to detect the reversal effect of Changweiqing on HCT-8 / V cell line. The expression of P-glycoprotein (P-gp) was determined by flow cytometry. Results: The resistance index of HCT-8 / V cell line to VCR, DDP, 5-FU and MMC were 18.6, 22.3, 4.3 and 3.7 respectively. After 48 hours of treatment, the drug resistance of Chemotherapy was significantly decreased, and the reversal indexes of VCR and DDP were 3.3 and 7.5, respectively. The expression of P-gp in HCT-8 / V cells was significantly higher at rest HCT-8 (P <0.01), while the expression of P-gp was significantly reduced by the serum of gastrointestinal drug (P <0.01). Conclusion: Changwei Decoction can reverse multidrug resistance of human colorectal cancer cell line HCT-8 / V, which may be related to its down-regulation of P-gp expression.
