目的 :观察在培养条件下血小板活化因子 (PAF)能否直接损伤原代人血管内皮细胞 (HVEC)和ECV - 30 4细胞系。方法 :利用光镜和结晶紫比色的方法 ,观察HVEC、ECV - 30 4细胞系与PAF孵育前后形态和生物活性变化。结果 :PAF与HVEC孵育 30min即发生细胞形态改变 ,生物活性则未见显著下降 ,与ECV - 30 4细胞系孵育后的细胞形态和生物活性未见明显变化。结论 :在培养条件下PAF可致HVEC损伤 ,对ECV - 30 4细胞系则未见明显损伤。
AIM: To observe whether platelet activating factor (PAF) can directly damage primary human vascular endothelial cells (HVEC) and ECV - 30 4 cell lines under culture conditions. Methods: The morphological and biological activities of HVEC, ECV - 30 4 cell lines before and after incubation with PAF were observed by light microscopy and crystal violet colorimetry. Results: After the cells were incubated with HVEC for 30 minutes, the cell morphology was changed and the biological activity was not significantly decreased. There was no obvious change in cell morphology and biological activity after incubated with ECV - 30 4 cell line. CONCLUSION: PAF can induce HVEC injury under culture conditions and no obvious damage to ECV - 30 4 cell line.
